ANTIGEN RECOGNIZED IN SPECIES (tested so far): Xenopus laevis, X. borealis, XLKE-A6 cells; human (see below). Negative with Pleurodelis, Triturus, Rana, chicken, rat, mouse
CLINICAL APPLICATION/SPECIFICITY: The antibody stains specifically the nucleolus and Cajal bodies
in therphase cells. The antigen is a constitutively expressed nucleolar component and a specific marker for the dense fibrillar component of the interphase nucleolus. The protein recognizes is a 180 kD polypeptide in Xenopus (xNopp180), homologue to the rat polypeptide (Nopp140). When applied at high concentration to human cells the antibody reacts with a nucleolar component which, however, does not correspond to xNopp180.
APPLICATION: Immunoblotting (western)
Immunoprecipitation
Immunofluorescence microscopy on frozen sections (also after fixation with paraformaldehyde)
WORKING DILUTION: 1:5 to 1:10 for Immunofluorescence
1:50 for immunoblotting (western, ECL method)
INCUBATION TIME: 1 h at room temperature
STORAGE: 2-8°C for immediate use or -20°C (aliquot)
