ANTIGEN
PREPARATION: Five different serotypes of Group A rotavirus strains WA (serotype I), DS-1 (serotype II), Ito (serotype III), HOCHI (serotype IV) and 69M (serotype VIII) were propagated on monolayers of MA monkey kidney cells. Each serotype was then purified by high speed centrifugation at 100,000 for 90 minutes through a sucrose cushion. The genomicRNA of each preparation was assessed using PAGE and its group, sub-group and serotype assessed by enzyme linked immunoassays based on a total of eleven different specific monoclonal antibodies. Thus each serotype was confirmed before immunization. Immunization was performed with live rotavirus serotypes to avoid the risk of damaging their surface viral proteins (VP7, VP6, VP4).
