DESCRIPTION: The predominant form of the Rubella antigen in this preparation is whole virions suspended in NTE with sucrose.
STRAIN: HPV-77
FORM: Highly purified antigen, in NTE Buffer (Sodium Chloride, Tris, and EDTA)
PROTEIN/CONCENTRATION: 0.42 mg/ml determined by dye binding assay.
PREPARATION: Vero cells infected with Rubella release virus particles into the culture supernatant. Supernatant is harvested and clarified by microfiltration. The material is then concentrated 200 fold by crossflow ultrafiltration. The Rubella antigen is then purified by two stages of sucrose density gradient centrifugation. The antigen suspension is adjusted to ensure batch to batch reproducibility.
APPLICATION: Immunoassay (ELISA, latex formats), sonicated immediately prior to use to ensure the uniformity of the antigen.
STORAGE TEMPERATURE: -70°C, Avoid repeated Freeze/Thaws
INACTIVATION: Rubella K2S antigen is inactivated by exposing the material to UVlight. This method was chosen to maximize loss of infectivity with a minimum loss of antigenicity.
