STRAIN: RH, cultured in Swiss Webster Mice
FORM: Soluble toxoplasma components in PBS
PURIFICATION: Toxoplasma organisms are harvested by centrifugation, washed in PBS and extracted using ether. Residual ether is removed from aqueous phas e by bubbling with nitrogen gas. The resulting antigen preparation contains soluble toxoplasma components in PBS. This antigen preparation contains a high concentration of disrupted tachyzoites in PBS without detergent or preservative. Some residual biological components from mouse ascites are present.
CONCENTRATION: 1 mg/ml protein
APPLICATION: Immunoassay, sonicated immediately prior to use to ensure the uniformity of the antigen
BUFFER: PBS, pH 7.2
SAFETY: The antigens have been inactivated by ether inactivation.
Use standard laboratory precautions before handling this material.
STORAGE: -20° - 70°C, Avoid repeated Freeze/Thaws
INACTIVATION: This preparation of Toxoplasma gondii tachyzoites is inactivated by disruption of cell membranes with sonication and with gamma irradiation. The gamma procedure works primarily by damaging genetic material.
Assay: The effectiveness of inactivation is tested by inoculating mice with the antigen preparation. The mice
are observed for 7 days for disease symptoms. The fluid contents of the peritoneum are examined for toxoplasma tachyzoites. If none are found a second passage and examination is performed. If no disease
symptoms are observed and toxoplasma is not found in the peritoneum, the preparation is considered inactivated.
