Background:
The reactivity of the antiserum is restricted to C1 esterase inhibitor. In immunoelectrophoresis and dial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified C1 esterase inhibitor. No reaction is obtained with any other plasma protein component or serum. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of C1 esterase inhibitor in human plasma or other body fluids or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included. In immunoelectrophoresis use 2 ?l or equivalent against 120 ?l antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 ?l in a 3 mm diameter center well and 2 ?l serum samples (neat and serially diluted) in 2 mm diameter peripheral wells.

Isotype: Rabbit Polyclonal

Source: C1 esterase inhibitor is a single chain acid glycoprotein with a molecular weight of 110,000. The protein has a high carbohydrate content. In immunoelectrophoresis it ha an alpha mobility. The concentration in normal serum ranges from 150 to 350 ?g/ml. Reduced levels are observed in hereditary angioedema. It regulates the activation of complement factor C1 as well as the activity of activated C1 by coupling with the active catalytic site at the light chains of C1r and C1s. Moreover it also inhibits, kallikrein, Factor XI and Factor XII. Freund's complete adjuvant is used in the first step of the immunization procedure.

Form: Delipidated, heat inactivated, in PBS and 0.1% sodium azide.

Reactivity: human

Cross Reactivity:
The antiserum does not cross react with any other component of Human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.

Applications: Immunoprecipitation

Storage: Store at 4C, or in small aliquots at -20C.
