Escherichia coli J5 (Rc mutant) (ATCC) was grown in nutrient agar. Culture was centrifuge, washed and the precipitated bacteria were suspended in isotonic saline at 1010 cfu./ml, aliquoted, and sonicated, using Ultra sonic bath. After centrifugation, the pellet was collected and used for immunization.
