Background:
Human GFAP-delta, also termed GFAP-?, is an alternative splice variant of the GFAP protein. GFAP proteins are selectively loCated in astrocytes and represent the major constituent of astrocytic intermediate filaments. GFAP expression levels are highly variable during development of the central nervous system. GFAP-delta differs from the predominant splice form of GFAP, GFAP-alpha, in its C-terminal sequence in which the original amino acid sequence, encoded by exons 8 and 9, are replaced by a unique and conserved tail of 42 amino acids encoded by exon 7+ and called the GFAP-delta tail domain. This GFAP-delta tail domain is able to bind presenilin proteins of which mutated forms are a cause of Alzheimer disease. GFAP-delta does however not discriminate between Alzheimer and control tissue. Upregulation of GFAP-delta has a negative effect on GFAP filament formation. In contrast to GFAP-alpha, GFAP-delta is not upregulated in astrogliosis.
