Introduction:
Alpha-fetoprotein (AFP, α-fetoprotein; also sometimes called alpha-1-fetoprotein, alpha-fetoglobulin, or alpha fetal protein) is a protein[5][6] that in humans is encoded by the AFP gene.[7][8] The AFP gene is located on the q arm of chromosome 4 (4q13.3).Maternal AFP serum level is used to screen for Down syndrome, neural tube defects, and other chromosomal abnormalities.AFP is a major plasma protein produced by the yolk sac and the fetal liver during fetal development. It is thought to be the fetal analog of serum albumin. AFP binds to copper, nickel, fatty acids and bilirubinand is found in monomeric, dimeric and trimeric forms.
Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-AFP polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-AFP polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the AFP amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of AFP can be calculated.

For quantitative detection of AFP in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
