Introduction:
Tumor necrosis factor receptor superfamily, member 4 (TNFRSF4), also known as CD134 and OX40 receptor, is a member of the TNFR-superfamily of receptors which is not constitutively expressed on resting naïve T cells, unlike CD28. OX40 is a secondary co-stimulatory immune checkpoint molecule, expressed after 24 to 72 hours following activation; its ligand, OX40L, is also not expressed on resting antigen presenting cells, but is following their activation. Expression of OX40 is dependent on full activation of the T cell; without CD28, expression of OX40 is delayed and of fourfold lower levels.

Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-CD134 / TNFRSF4 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-CD134 / TNFRSF4 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CD134 / TNFRSF4 amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of CD134 / TNFRSF4 can be calculated.

For quantitative detection of CD134 / TNFRSF4 in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.

Store at 2-8℃ for 6months, or at -20℃for 12 months.
