Human FGF21 ELISA Kit

Introduction:
Fibroblast growth factor 21 (FGF-21) is a protein that in mammals is encoded by the FGF21 gene.The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family and specifically a member of the endocrine subfamily which includes FGF23 and FGF15/19. FGF21 is the primary endogenous agonist of the FGF21 receptor, which is composed of the FGF receptor and co-receptor β-Klotho.FGF family members possess broad mitogenic and cell survival activities and are involved in a variety of biological processes including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion.[6] FGFs act through a family of four FGF receptors. Binding is complicated and requires both interaction of the FGF molecule with an FGF receptor and binding to heparin through a heparin binding domain.

Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-FGF21 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-FGF21 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the FGF21 amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of FGF21 can be calculated.

For quantitative detection of FGF21 in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.

Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
