Introduction:
Interleukin 13 (IL-13) is a protein that in humans is encoded by the IL13 gene.IL-13 was first cloned in 1993 and is located on chromosome 5q31.1 with a length of 1.4kb.It has a mass of 13 kDa and folds into 4 alpha helical bundles.The secondary structural features of IL-13 are similar to that of Interleukin 4 (IL-4); however it only has 25% sequence identity to IL-4 and is capable of IL-4 independent signaling.IL-13 is a cytokine secreted by T helper type 2 (Th2) cells, CD4 cells, natural killer T cell, mast cells, basophils, eosinophils and nuocytes.Interleukin-13 is a central regulator in IgE synthesis, goblet cell hyperplasia, mucus hypersecretion, airway hyperresponsiveness, fibrosis and chitinase up-regulation.It is a mediator of allergic inflammation and different diseases including asthma, and atopic dermatitis.
Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-13 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-IL-13 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-13 amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of IL-13 can be calculated.
Application:
For quantitative detection of IL-13 in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
