Introduction:
Interleukin-17A is a protein that in humans is encoded by the IL17A gene. In rodents, IL-17A used to be referred to as CTLA8, after the similarity with a viral gene (O40633).The protein encoded by this gene is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO).

Principle of the Assay:
Incubate the acceptor beads coated with anti-IL-17A antibody 1,biotin conjugated anti-IL-17A antibody 2,and the sample to form a sandwich immune complex ( antibody 1-sample- antibody 2). the immune complex is the n incubated withdonor beads coated with streptavid into form a chemiluminescent complex, ensuring that the distance between the two beads is less than 200 nm. When the distance between the two beads is less than 200 nm, the donor beads is excited by light (680 nm), producing singlet oxygen which diffuses to the acceptor beads, inducing energy transfer. the acceptor beads absorb this energy and emit scattering light at 615 nm. the light signal is collected by a photosensitive detector, and the concentration ofIL-17Ain the sample is calculated through mathe matical fitting. In the absence of humanIL-17Ain the sample, no immune complex is formed, or the distance between the two beads exceeds 200 nm, exceeding the transfer distance for singlet oxygen, and the acceptor beads do not emit any light.

Application:
For quantitative detection of human IL-17A in buffer solution, serum, cell culture supernatant.
