Introduction:
Interleukin 5 (IL-5) is an interleukin produced by type-2 T helper cells and mast cells.Through binding to the interleukin-5 receptor, interleukin 5 stimulates B cell growth and increases immunoglobulin secretion—primarily IgA. It is also a key mediator in eosinophil activation. IL-5 is a 115-amino acid (in human, 133 in the mouse) -long Th2 cytokine that is part of the hematopoietic family. Unlike other members of this cytokine family (namely interleukin 3 and GM-CSF), this glycoprotein in its active form is a homodimer.
Principle of the Assay:
Incubate the Fconjugated anti-IL-5 antibody 1,biotin conjugated anti-IL-5 antibody 2,acceptor beads coated with F-conjugate,and the sample to form a sandwich immune complex ( antibody 1-sample- antibody 2). the immune complex is the n incubated withdonor beads coated with streptavid into form a chemiluminescent complex, ensuring that the distance between the two beads is less than 200 nm. When the distance between the two beads is less than 200 nm, the donor beads is excited by light (680 nm), producing singlet oxygen which diffuses to the acceptor beads, inducing energy transfer. the acceptor beads absorb this energy and emit scattering light at 615 nm. the light signal is collected by a photosensitive detector, and the concentration ofIL-5in the sample is calculated through mathe matical fitting. In the absence of humanIL-5in the sample, no immune complex is formed, or the distance between the two beads exceeds 200 nm, exceeding the transfer distance for singlet oxygen, and the acceptor beads do not emit any light.
Application:
For quantitative detection of human IL-5 in buffer solution, serum, cell culture supernatant.
Storage and Expiration:
Store at 2-8℃ for 6 months.
