Introduction:
Macrophage Inflammatory Protein-3 (MIP-3α), also known as Chemokine (C-C motif) ligand 20 (CCL20) or liver activation regulated chemokine (LARC), is a small cytokine belonging to the CC chemokine family. the CCL20 gene contains 4 exons and maps to chromosome 2q35-q36. It is strongly chemotactic for lymphocytes and weakly attracts neutrophils. CCL20 is expressed in several tissues with highest expression observed in peripheral blood lymphocytes, lymph nodes, liver, appendix, and fetal lung and lower levels in thymus, testis, prostate and gut. CCL20 is implicated in the formation and function of mucosal lymphoid tissues via chemoattraction of lymphocytes and dendritic cells towards the epithelial cells surrounding these tissues. CCL20 elicits its effects on its target cells by binding and activating the chemokine receptor CCR6.
Principle of the Assay:
This kit was based onsandwich enzyme-linked immune-sorbent assay technology. Anti-MIP-3αpolyclonalantibodywaspre-coated onto 96-well plates. And thebiotin conjugatedanti-MIP-3αpolyclonal antibody wasused as detection antibodies. Thestandards,test samples andbiotinconjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washedaway with wash buffer. TMB substrateswereused to visualize HRP enzymatic reaction. TMB was catalyzed byHRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to theMIP-3αamount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MIP-3αcan be calculated.
Application:
For quantitative detection of MIP-α in human serum, plasma, body fluids, tissue lysates or cell culture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
