Introduction:
Granulin is a protein that in humans is encoded by the GRN gene.Each granulin protein is cleaved from the precursor progranulin, a 593 amino-acid-long and 68.5 kDa protein.[8] While the function of progranulin and granulin have yet to be determined, both forms of the protein have been implicated in development, inflammation, cell proliferation and protein homeostasis. The 2006 discovery of the GRN mutation in a population of patients with frontotemporal dementia has spurred much research in uncovering the function and involvement in disease of progranulin in the body. While there is a growing body of research on progranulin's role in the body, studies on specific granulin residues are still limited.
Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-progranulin polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-progranulin polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the progranulin amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of progranulin can be calculated.
Application:
For quantitative detection of progranul in in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
