Introduction:
Tenascin C (TN-C) is a glycoprotein that in humans is encoded by the TNC gene.It is expressed in the extracellular matrix of various tissues during development, disease or injury, and in restricted neurogenic areas of the central nervous system.Tenascin-C is the founding member of the tenascin protein family. In the embryo it is made by migrating cells like the neural crest; it is also abundant in developing tendons, bone and cartilage.The human tenascin C gene, TN-C, is located on chromosome 9 with location of the cytogenic band at the 9q33. The entire Tenascin family coding region spans approximately 80 kilobases translating into 2203 amino acids.
Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Tenascin-C polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Tenascin-C polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Tenascin-C amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of Tenascin-C can be calculated.
Application:
For quantitative detection of Tenasc in -C in human serum, plasma, body fluids, tissue lysatesor cellculture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
