Datasheet
Background:
The NF-κB/Rel transcription factors are present in the cytosol in an inactive state, complexed with the inhibitory IκB proteins. Most agents that activate NF-κB do so through a common pathway based on phosphorylation-induced, proteasome-mediated degradation of IκB. The key regulatory step in this pathway involves activation of a high molecular weight IκB kinase (IKK) complex whose catalysis is generally carried out by three tightly associated IKK subunits. IKKα and IKKβ serve as the catalytic subunits of the kinase and IKKγ serves as the regulatory subunit. Activation of IKK depends upon phosphorylation at Ser177 and Ser181 in the activation loop of IKKβ (Ser176 and Ser180 in IKKα), which causes conformational changes, resulting in kinase activation.

Alternative Name:
I kappa-B kinase alpha, IkBKA, IKK-alpha, IKK-A, IkappaB kinase, EC 2.7.11.10, I-kappa-B kinase 1, IKK1, Conserved helix-loop-helix ubiquitous kinase, Nuclear factor NF-kappa-B inhibitor kinase alpha, NFKBIKA, CHUK, IKKA, TCF16

Application Dilution: WB: 1:500~1:1000, IHC: 1:50~1:200

Specificity: p-IKKα/β (S180/181) polyclonal antibody detects endogenous levels of IKKα only when phosphorylated at Ser180 and IKKβ only when phosphorylated at Ser181.

Immunogen:
Synthetic phosphopeptide derived from human IKKα/β around the phosphorylation site of Serine 180/181.

MW: ~ 85 kDa (IKKα); 87kDa (IKKβ)

Swis Prot.: O15111/O14920

Purification & Purity:
The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen and the purity is > 95% (by SDS-PAGE).

Format:
1 mg/ml in Phosphate buffered saline (PBS) with 0.05% sodium azide, approx. pH 7.2.

Storage:
Store at 4°C short term. Aliquot and store at -20°C long term. Avoid freeze-thaw cycles.
