Background:
Integrins are cell adhesion molecules that can mediate bidirectional transfer of signals across the plasma membrane. Integrin α6β4receptors are found in basement membrane along with laminin-5. These receptors are expressed in epithelial, schwann, endothelial and some immune cells. The cytoplasmic domain of the Integrin β4 subunit recruits the adaptor protein Shc and is required for assembly of hemidesmosomes. Tyrosine phosphorylation of multiple sites within the cytoplasmic domain regulates these cellular events. In particular, Tyr-1526 interacts with the phosphotyrosine binding domain of Shc and is required for Shc activation. In addition, Tyr-1494 is required for integrin-mediated IRS-2 phosphorylation and activation of PI3-kinase. More importantly this site is critical for integrin α6β4 increases in carcinoma invasion. Human recombinant Integrin β4 (Cytoplasmic) protein is a 23 kDa fragment that includes amino acids 1619 to 1822 plus a C-terminal His-tag. This integrin β4 protein is detected by mouse monoclonal anti-Integrin β4 (Cat.#IM1261).
Buffer/Storage:
Integrin β4 recombinant protein lysate is supplied at a concentration of 2 ng/µl in electrophoresis sample buffer (62.5 mM Tris pH 6.8, 2% SDS, 5% glycerol, 0.003% bromophenol blue, 0.9% β-mercaptoethanol). Store at –20°C. Do not boil or dilute. Stable for 1 year.
