Introduction:
Stromelysin-1 also known as matrix metalloproteinase-3 (MMP-3) is an enzyme that in humans is encoded by the MMP3 gene. The MMP3 gene is part of a cluster of MMP genes which localize to chromosome 11q22.3.MMP-3 has an estimated molecular weight of 54 kDa.The MMP-3 enzyme degrades collagen types II, III, IV, IX, and X, proteoglycans, fibronectin, laminin, and elastin. In addition, MMP-3 can also activate other MMPs such as MMP-1, MMP-7, and MMP-9, rendering MMP-3 crucial in connective tissue remodeling. The enzyme is also thought to be involved in wound repair, progression of atherosclerosis, and tumor initiation.
Principle of the Assay:
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-MMP-3 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MMP-3 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MMP-3 amount of sample captured in plate. Read the O. D. absorbance at 450nm in a microplate reader, and then the concentration of MMP-3 can be calculated.
Application:
For quantitative detection of MMP-3 in mouse serum, plasma, body fluids, tissue lysatesor cellculture supernatants.
Storage and Expiration:
Store at 2-8℃ for 6months, or at -20℃for 12 months.
