The encoded protein functions as a transcriptionalrepressor during embryogenesis through interactions with componentsof the core transcription complex and other homeoproteins. It mayalso have roles in limb-pattern formation, craniofacialdevelopment, particularly odontogenesis, and tumor growthinhibition. Mutations in this gene, which was once known ashomeobox 7, have been associated with nonsyndromic cleft lip withor without cleft palate 5, Witkop syndrome, Wolf-Hirschom syndrome,and autosomoal dominant hypodontia.
