Description: Mouse IgG monoclonal antibody for p27Kip1, cyclin-dependent kinase inhibitor 1B (p27, Kip1) (CDKN1B) detection. Tested with WB, ICC in Human;mouse;rat. No cross reactivity with other proteins.Protein Name: Cyclin-dependent kinase inhibitor 1B
Protein Function:
CDKN1B: Important regulator of cell cycle progression. Involvedin G1 arrest. Potent inhibitor of cyclin E- and cyclin A-CDK2complexes. Forms a complex with cyclin type D-CDK4 complexes andis involved in the assembly, stability, and modulation of CCND1-CDK4 complex activation. Acts either as an inhibitor or anactivator of cyclin type D-CDK4 complexes depending on itsphosphorylation state and/or stoichometry.
Clone Name(s):
DCS-72
Subcellular Localization: CDKN1B: Nucleus. Cytoplasm. Endosome Note: Nuclear and cytoplasmic in quiescent cells. AKT- or RSK-mediated phosphorylation on Thr-198, binds 14-3-3, translocates tothe cytoplasm and promotes cell cycle progression. Mitogen-activated UHMK1 phosphorylation on Ser-10 also results intranslocation to the cytoplasm and cell cycle progression.Phosphorylation on Ser-10 facilitates nuclear export. Translocatesto the nucleus on phosphorylation of Tyr-88 and Tyr-89.Colocalizes at the endosome with SNX6; this leads to lysosomaldegradation (By similarity). |Cdkn1b: Nucleus. Cytoplasm. Endosome. Note: Nuclearand cytoplasmic in quiescent cells. AKT- or RSK-mediatedphosphorylation on Thr-197, binds 14-3-3, translocates to thecytoplasm and promotes cell cycle progression. Mitogen-activatedUHMK1 phosphorylation on Ser-10 also results in translocation tothe cytoplasm and cell cycle progression. Phosphorylation on Ser-10 facilitates nuclear export. Translocates to the nucleus onphosphorylation of Tyr-88 and Tyr-89 (By similarity). Colocalizesat the endosome with SNX6; this leads to lysosomal degradation.
Tissue Specificity: CDKN1B: Expressed in all tissues tested. Highestlevels in skeletal muscle, lowest in liver and kidney.
Immunogen: recombinant rodent p27Kip1 protein.
Calculated MW: 22139 MW
Purification: Ascites
Format: Lyophilized
Reconstitution: Add 1ml of PBS buffer will yield a concentration of 100ug/ml.
Contents: Mouse ascites fluid, 1.2% sodium acetate, 2mg BSA, with 0.01mg NaN3 as preservative.
Concentration (mg/ml): Add 1ml of PBS buffer will yield a concentration of 100ug/ml.
Storage: At -20˚C for one year. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for a longer time.Avoid repeated freezing and thawing.
