Background:
PDGF is amitogen formesenchyme- and glia-derived cells. Itconsists of two disulfide-bonded polypeptide chains, A and B, and occurs asthree isoforms, PDGF-AA, PDGF-AB and PDGF-BB. The three isoforms bindwith different affinitiesto two receptor types, A and B,which are structurallyrelated and possess protein-tyrosine kinase domains.Ligand binding induces activation of the receptorkinases byformation of receptor dimers; the A subunit of PDGF binds onlyto A receptorswith high affinity,whereasthe B subunitcan bind to both A and B receptors. Evidence suggeststhat PDGFmayfunction as a neurotrophicfactor.
