Protein Family: Enzymes, Protein Kinases

Pathway and Disease: Cell Growth and Death, Metabolic Disorders, Translation

Alternate Names: Eukaryotic translation initiation factor 2-alpha kinase 3; PRKR-like endoplasmic reticulum kinase; Pancreatic eIF2-alpha kinase; HsPEK; EIF2AK3; PEK; PERK

Accession No.: Q9NZJ5, NP_004827

Description:
PERK, a Ser/Thr protein kinase, phosphorylates the alpha subunit of eukaryotic translation-initiation factor 2 (EIF2), leading to its inactivation and to a rapid reduction of translational initiation and repression of global protein synthesis. PERK serves as a critical effector of unfolded protein response (UPR)-induced G1 growth arrest due to the loss of cyclin D1. Defects in PERK are the cause of Wolcott-Rallison syndrome, an early-onset diabetes mellitus.

Format:
Each vial contains 0.1 mg of lyophilized peptide. Reconstitute with 0.1 ml deionized water for a final concentration of 1 mg/ml. Use at 5.6 ug/ml for a 100X excess over antibody for maximum blocking effect.

MW: 2061.2 g/mol

Sequence:
The synthetic peptide used to raise the antibody Cat. No. 200117 is selected from a sequence within the C-term region of human PERK. For blocking experiments, a 10 to 100 fold molar excess to antibody is recommended.

Composition:
C79H141N35O28S1

Purity:
> 80% by HPLC

Solubility:
Distilled water for a solution up to 2 mg/ml, otherwise we recommend using acetonitrile.

Storage:
Store at -20°C. The product is hygroscopic and must be protected from light. Product is guaranteed one year from the date of shipment. Following reconstitution, store at -20°C.
