Background:
One family of inhibitory axon guidance molecules is the semaphorins. The semaphorins include secreted, transmembrane, and GPI-anchored extracellular molecules that have been implicated in neuron development, vascular disease, and tumor progression. There are eight classes of semaphorin genes, all of which are characterized by a conserved 500 amino acid, cystine-rich Sema domain. Semaphorin 3A (Sema-3A) binds its co-receptors, Neuropilin-1 and Plexin A1, and this interaction has been implicated in suppressing axon regeneration after injury. Sema-3E binds Plexin D1 receptor, and has putative roles in axon guidance, angiogenesis, and cancers. The Sema-3E protein is expressed as full length (87 kDa) and furin-dependent processed forms (61 and 25 kDa). Sema-3F binds with high affinity to Neuropilin-2, and low affinity to Neuropilin-1. Sema-3F has been implicated in neuronal development, angiogenesis, and tumor progression. Sema-4D binds Plexin B1, and this pathway may be involved in cell survival and angiogenesis.
Buffer/Storage:
Rabbit polyclonal antibodies are supplied in phosphate-buffered saline, 50% glycerol, 1 mg/ml BSA, and 0.05% sodium azide. Store all at –20°C. Stable for 1 year.
