Background:
Transcription of protein-coding genes can be reconstitutedon naked DNA with only the general transcription factors and RNApolymerase II. However, this minimal system cannot transcribe DNApackaged into chromatin, indicating that accessory factors mayfacilitate access to DNA. One such factor, FACT (facilitateschromatin transcription), interacts specifically with histonesH2A/H2B to effect nucleosome disassembly and transcriptionelongation.
