Thrombopoietin (THPO) is a lineage-specific cytokine affecting proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development, and is a major physiological regulator of circulating platelets. Recombinant Human Thrombopoietin produced in E. coli is a single non-glycosylated polypeptide chain containing 174 amino acids with a MW of 18,608 Da which comprises the receptor binding domain of the Mpl-ligand protein.

Application Notes:
Recombinant TPO is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of MO7e cells is less then 0.8 ng/ml.

Purity > 98% as determined by analysis by RP-HPLC and reducing and non-reducing SDS-PAGE. Protein Content determined by UV spectroscopy at 280 nm and analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard.
Format:
Each vial contains 10 ug protein lyophilized in 10 mM sodium phosphate pH8.0. Reconstitute with 0.1 ml sterile deionized water for a final concentration of 0.1 mg/ml.
Storage: Store at -20°C. Product is guaranteed one year from the date of shipment. Following reconstitution, store at -20°C. We recommend to add a carrier protein (0.1% HSA or BSA) for long term storage.
