Tumor necrosis factor-alpha (TNF-alpha) is a cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by mactrophages and can induce cell death of certain cell lines. It is a potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia. Recombinant mouse TNF-alpha produced in S. lividans is a single non-glycosylated polypeptide chain containing 156 amino acids with a MW of 17301.32 Da.

Source: Streptomyces lividans. Endotoxin level as measured by LAL is <0.01ng/ug or <0.1EU/ug.

Application Notes:
rmTNF-alpha is fully biologically active when compared to standard. The ED50 as determined by the cytolysis of mouse L929 cells in the presence of Actinomycin D is less then 0.05ng/ml, corresponding to a Specific Activity of 9x108 IU/mg.

Purity > 98% as determined by RP-HPLC and reducing and non-reducing SDS-PAGE. Protein Content determined by UV spectroscopy at 280 nm and analysis by RP-HPLC calibrated against a known standard. Quantitation on SDS-PAGE against a known standard.

Format:
Each vial contains 20 ug protein lyophilized in 50 mM Tris-HCl, pH8.0, 20 mM NaCl. Reconstitute with 0.2 ml sterile deionized water for a final concentration of 0.1 mg/ml.

Storage: Store at -20°C. Product is guaranteed one year from the date of shipment. Following reconstitution, store at -20°C. We recommend to add a carrier protein (0.1% HSA or BSA) for long term storage.
